Yuxin An

From September 2016 to January 2023, Dr. An pursued doctoral studies in Analytical Chemistry at the Dalian Institute of Chemical Physics (DICP), Chinese Academy of Sciences (CAS), earning a Ph.D. degree. Since August 2023, she has been serving as a Technologist-in-charge at the Zhejiang Provincial Center for Disease Control and Prevention. To address the challenges posed by the high dynamics, low abundance, and high spatial heterogeneity of protein complexes, she has carried out a series of research efforts aimed at achieving precise in situ characterization of protein complexes. She developed an organelle-targeted method for in situ analysis of protein complexes, enabling precise mapping of their conformations and interaction interfaces within specific subcellular compartments. She further advanced a deep-coverage analysis strategy for in situ structural characterization of protein complexes based on the removal of dead-end cross-linked peptides, achieving efficient enrichment of cross-linked peptides and significantly improving cross-linking identification coverage. In addition, she developed a novel chemical cross-linking agent featuring membrane permeability, enrichability, and high biocompatibility, and established an in situ chemical cross-linking analysis method in living cells. This approach greatly improved the coverage and accuracy of characterizing weakly bound, readily dissociable, and low-abundance protein–protein interactions, as well as the structural analysis of intrinsically disordered regions within proteins. Collectively, these efforts provide critical technical support and an essential data foundation for comprehensively elucidating the biological functions of proteins.
